They’re stuffing runs early, but allowing Seahawks RBs to wiggle away and find yards. – The 49ers are just doing a poor job of tackling. – Good work by WR Danny Gray on special teams to help pin Seattle inside their own 20 after a nice punt by Mitch Wishnowsky. – Another bad throw from Purdy on third-and-3 and the 49ers will punt. Seattle is in the backfield on every play. – The interior of the 49ers’ offensive line is having an abysmal first half. A long drive and any points would be huge because the 49ers get the ball back after halftime. – 5:01 left with the 49ers offense taking the field down a point. This is what happens when red zone trips turn into field goal instead of touchdowns. – Deep shot to DK Metcalf goes for a 50-yard TD and Seattle has the lead. This is as off-balance as the 49ers have looked on defense since the Chiefs game. – Seattle is killing San Francisco with play action. – Another big return puts Seattle at their own 30 to start their drive after the 49ers’ field goal. That sets up third-and-17 and the 49ers stall out in the red zone again. – Purdy took a sack on a blown up screen where Aaron Banks got worked on the interior. Expecting plenty of play action as this game plays out. – Seattle is getting downhill so fast against the run. Samuel was wide open on a crossing route. Purdy tries a shot to Aiyuk and it’s nearly intercepted. On second-and-12 he scrambled for 13 yards and a first down. – The Seahawks are selling out to stop the run. Purdy hits Aiyuk for 23 yards on a play action boot. – Good response by the 49ers after Seattle’s TD. Bad tackling and undisciplined against the run and pass. The 49ers defense looked terrible on that series. They ran a direct snap to Walker after Smith lined up under the right guard. The longer they stay in this game the harder they’ll be to defend. Seattle is dedicated to running the ball here. – Fred Warner and Charles Omenihu were both shaken up on a Walker carry early in the second quarter. RB Kenneth Walker is a heck of a player.Įnd of first quarter: 49ers 10, Seahawks 0 – Seattle’s offense is starting to roll a little bit late in the first quarter. – Deommodore Lenoir matched up on DK Metcalf is not an advantageous matchup for the 49ers. Second-and-goal, nobody is open so he dances around in the pocket and found a wide open Christian McCaffrey for an easy touchdown. – Christian McCaffrey’s first touch is a handoff that goes for 66 yards. Geno Smith kept it, but Tashaun Gipson made an unbelievable play to track him down for a loss and force another Seattle punt. – Another third-and-2 for the Seahawks and they try a read option. That’s the kind of thing that will keep them in the game. – Seattle starts their second drive at their own 38 thanks to a big return. Robbie Gould hit a short field goal to put the 49ers up 3-0 early. He’s missed a couple throws down the field, and on third-and-3 in the red zone he was nearly intercepted after what looked like maybe a miscommunication with Jauan Jennings, who he badly underthrew earlier in the drive. Good bounce-back play from the rookie QB. McCaffrey motioned out and Purdy hit Brandon Aiyuk for 19 yards. – Samuel and Christian McCaffrey in the backfield on second-and-10. He missed way too high and nearly threw an interception. Brock Purdy’s first throw was a shot to Deebo Samuel and it was not good. The Seahawks tried blocking him one-on-one, and that was not going to work. – Arik Armstead comes up with a sack on third-and-2! Great work from him against Seattle’s left guard Gabe Jackson. – Nice stop by Javon Kinlaw on a second-and-short on Seattle’s opening series.

Treat the dsDNA fragment that you are using as a probe with a limiting amount of Dnase, which causes double-stranded nicks in DNA. Biotin/streptavidin detection is done by colorimetric methods, and bioluminescent visualization uses luminesence. If you used a radiolabeled 32P probe, then you would visualize by autoradiograph. Visualize your radioactively labeled target sequence. To hybridize, use the same buffer as for prehybridization, but add your specific probe.Ħ. Probing is often done with 32P labeled ATP, biotin/streptavidin or a bioluminescent probe.Ī prehybridization step is required before hybridization to block non-specific sites, since you don't want your single-stranded probe binding just anywhere on the membrane. Whatever you call it, this process relies on the ssDNA hybridizing (annealing) to the DNA on the membrane due to the binding of complementary strands. (You can also bake nitrocellulose at about 80C for a couple of hours, but be aware that it is very combustible.)ĥ. This cross links (via covalent bonds) the DNA to the membrane. (SSC provides the high salt level that you need to transfer DNA.)Īfter you transfer your DNA to the membrane, treat it with UV light. This works similarly to capillary action, except more SSC goes through the gel and membrane, so it is faster (about an hour). In this procedure, a vacuum sucks SSC through the membrane. You may use a vacuum blot apparatus instead of capillary action. Capillary action transfer draws the buffer up by capillary action through the gel an into the membrane, which will bind ssDNA. Transfer is usually done by capillary action, which takes several hours. Many scientists feel nylon is better since it binds more and is less fragile. Nitrocellulose typically has a binding capacity of about 100µg/cm, while nylon has a binding capacity of about 500 µg/cm. Traditionally, a nitrocellulose membrane is used, although nylon or a positively charged nylon membrane may be used. Transfer the denatured DNA to the membrane. Be aware, however, that the procedure may also be hampered by fragments that are too small.īe sure to neutralize the acid after this step, or the base after the prior step if you don't depurinate.Ĥ. Depurination with HCl (about 0.2M HCl for 15 minutes) takes the purines out, cutting the DNA into smaller fragments. Fragments greater than 15 kb are hard to transfer to the blotting membrane. Only ssDNA can transfer.Ī depurination step is optional. Denature the DNA (usually while it is still on the gel).įor example, soak it in about 0.5M NaOH, which would separate double-stranded DNA into single-stranded DNA. Digest the DNA with an appropriate restriction enzyme.ģ. Let's look at this technique in greater detail.ġ. This diagram shows the basic steps involved in a Southern blot. Under optimal conditions, you can expect to detect 0.1 pg of the DNA for which you are probing. The amount of DNA needed for this technique is dependent on the size and specific activity of the probe. For example, Southern Blotting could be used to locate a particular gene within an entire genome. Southern blotting is designed to locate a particular sequence of DNA within a complex mixture. To oversimplify, DNA molecules are transferred from an agarose gel onto a membrane. Southern who developed this procedure at Edinburgh University in the 1970s. Southern blotting was named after Edward M. Each vignette takes about 20 minutes or so, and I honestly wish they’d been a little longer, but they were sweet. Waka’s World is made up of four short, but cute, stories starring our favorite manager’s various personalities from Memories: the okama, the general, the silent type, and the assassin. None of the routes were outright bad, but when looking at what they were supposed to follow up on, most of them didn’t leave much of an impact. Like with Shin, it felt almost like Ukyo underwent a character reset, tossing out any growth from the previous game to give what seemed like a forced conflict. Ukyo’s route fell somewhere in between for me and ended up being a bit disappointing, seeing as his route in Memories was honestly one of the best. This is one of those instances where a bit more spice would have made a lot of sense, though that’s a minor quibble. I loved Ikki and Kent’s routes, though I feel like the Teen rating held Ikki’s back a bit. Shin’s route felt like a character reset, while Toma’s didn’t address the whole cage thing nearly enough and fell sort of flat for me. For the most part, I found them a bit underwhelming, except for Ikki and Kent. Does Shin learn to be nicer to you? How does living together with Ikki turn out? Does Kent ever manage to make it to London? Exactly how does Toma recover from his actions? And how does Ukyo reconcile the multiple lives he’s lived with the one he’s got now? Unlike in Memories, there are no Bad Endings here and your choices don’t matter, as each route is basically glorified wish fulfillment with the lover of your choice. Know only that it is my favorite route across all three games, and I encourage you to try it out for yourselves!Īs this is a sequel, there will be spoilers for Amnesia: Memories going forward.Īfter Story is exactly what it says on the tin: Explore your relationship with the chosen love interest following the events of that fateful August. There’s also an unlockable fifth route if you complete all of the above, but it’s worth it to discover what that route is on your own. While on this outing, you’ll have the chance to select which character you’d like to spend time with, opening up three possible routes to explore: After Story, which continues each love interest’s story from Memories Waka’s World, a series of vignettes exploring the various personalities of the cafe’s manager and Girls Party, where the heroine and her friends chat about the boys. Everyone from Meido no Hitsuji, as well as Rika and Ukyo, want to take the heroine firefly hunting. The first of the fandisk games, Later, begins with a firefly outing set in a parallel New World from Memories. How do these two games stack up to the original, and are they worth your time? Let’s find out! Welcome back, Madams and Masters, to part two of my Amnesia review series! Last time I covered Amnesia: Memories, but today we’re going to dive deep into the fandisk sequels, Later and Crowd, released for the first time outside Japan in the Later x Crowd compilation for the Nintendo Switch. The goal was to create a family of fonts that appear at least distantly related. Some fonts, like Runic, are so obscure that typographers at Monotype built the font from scratch using stone engravings for inspiration. The Arabic typeface is emphasized in left to right strokes, while French's letters carry their weight in their vertical stems. The Tibetan script, for example, draws heavily from a calligraphic tradition, while English is more linear and geometric. And making those fonts “unmistakably Google” is nearly impossible. “That was always the complaint we got-fonts just don’t look good when you mix languages.” But developing a typeface for 800 languages that feels cohesive yet respectful of each language's cultural heritage created inherent tension. “You’d get this mishmash of fonts,” Jung says. So they’d buy one for Latin scripts like Spanish and French, another for Arabic scripts, and yet another for Japanese and Chinese. In the past, platforms licensed fonts for different scripts piecemeal (a script can cover a number of languages-English, Icelandic, and Dutch are just three of the dozens of languages that rely on Latin script). He led hundreds of researchers, designers, and linguists in giving every Unicode-enabled script a font, and ensuring those fonts feel like part of the same typographic family. If these less common languages have a digital typeface, it’s probably not well considered, because “certain regions of the world have much more rich typographic traditions than others,” says Steve Matteson, Monotype’s creative director. But people read and write in hundreds of languages. It is important because ALL galaxies emit red shift or waves which are stretched. What so important about the Doppler Shift? Try and get the same results using your calculator. The simulation also uses the same equation as above to work out the length of the stretched wavelength. To work out how long is the stretched wavelength considering time dilation, the relativistic factor is used and it is to be multiplied to the length of the overall stretched wavelength worked out by using the Doppler shift factor as shown below. The Doppler shift factor ignores time dilation so v has to be smaller than c for the factor to work. Notice v has to be much smaller than c because time dilation happens when approaching the speed of light, this is where the rate of time slows down so the waves rotating phasors rotate slower and so their wavelengths are stretched even more. The equation derived is know as the Doppler shift factor and it is used to work out how much the wavelength has been stretched. The factor of Doppler shift can be derived as shown below. Waves being stretched are known to undergo red shift as their wavelengths has increased towards the red end of the spectrum. The Doppler effect or Doppler shift is the result of a wave emitting source to move causing the wavelength of the wave to stretch or compress. The change colour button changes the colour scheme of the simulation from a selection of : The pause/play button will pause or unpause the simulation. The start/stop button will start or stop the simulation. The length of the stretched wavelength is as shown in diagram 1. It can be turned on or off in the drop down menu as shown in diagram 2. The value of the relativistic factor is as shown in diagram 1. The relativistic can be considered or ignored in this simulation by turning it on or off. The wavelength of the magnetic radiation is as shown in diagram 1. It can be edited as shown in diagram 2, click on it and press the number keys to change the variable. The speed of the source over the speed of light is as shown in diagram 1. This is where a part of the wave goes through one full phase rotation. The source of radiation is represented as a green spot. The status bar on the browser displays the frame rate the simulation is performing, a frame rate of 60 is considered satisfactory. The interface in this simulation consist of two sides, the left hand side shows the simulation and the right hand side shows the control panel. Because this is a model, the frequency of the wave front emitted is not to scale. Each wave front is represented as a line. This simulation models electromagnetic waves being emitted from a moving source. This browser does not have a Java Plug-in. In recent weeks they have laid siege to several provincial capitals across the country as the last US and Nato troops leave the country. Taliban fighters have swept through large swathes of Afghanistan at surprising speed, initially taking districts, many in remote areas. They requested to remain anonymous fearing retaliation from both sides. Heavy airstrikes were reported by residents of Sheberghan who also said the Taliban had freed prisoners from the city jail. Sheberghan is particularly strategic because it is the stronghold of US allied Uzbek warlord Rashid Dostum, whose militias are among those resurrected to aid the Afghan National Security and Defence Forces. On Friday, the Taliban took control of the southwestern Nimroz provincial capital of Zaranj, where the government says it is still battling insurgents inside the capital. Several other of the country's 34 provincial capitals are threatened. If the city falls, it will be the second provincial capital in as many days to succumb to the Taliban. The government did not deny politician Mohammad Karim Jawzjani's claim that Taliban fighters had entered Sheberghan, but said the city had not fallen. The final US military personnel are due to be withdrawn from Afghanistan on August 31, which has led to the Taliban offensive. Similarly, another clip on Twitter appeared to show something similar had happened in Zaranj, more than 700 miles away. It is the second provincial capital to fall to the insurgents over the past 24 hours after Zaranj fell yesterday.Ī video has emerged on social media which appears to show prisoners fleeing in Shiberghan after the Taliban managed to gain control of the prison. The Taliban have been advancing across Afghanistan after the US-led coalition pulled out of the war torn country, leaving the nation's struggling defence forces to deal with the terrorists.Įarlier today, the Taliban captured Sheberghan city in Jawzjan, the area's deputy governor confirmed to AFP. They are being supported by the AC-130 Spectre gunships which are armed with a 25mm Gatling gun, a 40mm Bofors cannon and a 105mm M102 cannon - which can provide pinpoint accurate fire from the air. The Cold War-era strategic bomber first flew in the 1950s but is still used due to its 70,000lb payload and range of more than 8,000 miles. This T-Shirt, new from The Warbird Collection, is sure to be a favorite for any aviation enthusiast, pilot, military aviator, flight crew and more.US President Joe Biden has ordered B-52 bombers and Spectre gunships to target Taliban terrorists in Afghanistan who are advancing towards three key cities. The Bella+Canvas shirts were selected for their premium feel, long lasting durability and excellent fit. This brand new T-Shirt design features the powerful AC-130 Spectre Special Operations Gunship aircraft, printed on a premium Bella+Canvas cotton shirt. Able to operate at night under the cover of darkness this massive aircraft carries a wide assortment of cannon, machine guns, and artillery and is able to rain sustained destruction down on enemy forces. The Spectre excels at supporting special operations troops, or any troops in contact with the enemy. The Lockheed C-130 Hercules can do a variety of missions, from airdropping cargo and troops to arial refueling and more, but perhaps the biggest modification to the iconic Herc comes in the form of the AC-130 Spectre Gunship.
